I russi hanno pero' bisogno di Indiana Jones (Harrison Ford) per trovare il reperto: cosi' costringono nella ricerca il famoso archeologo e il suo compagno Mac. Dopo essere riuscito a scappare, Indiana torna a casa, ma si ritrova preso di mira dall'FBI, che sospetta sia una spia russa. Come se non bastasse, il college dove lavora lo sospende dall'insegnamento. Deluso e amareggiato, decide di trasferirsi a Londra. Nel frattempo incontra il giovane Mutt (Shia LaBeouf) che gli chiede aiuto per ritrovare la madre, rapita dopo essersi interessata alla scomparsa del professor Oxley (John Hurt), il quale le aveva lasciato una lettera da decifrare. Indiana individua una pista che conduce a Nazca e parte con Mutt alla volta del Peru'. I due sono pero' inseguiti dagli agenti del KGB e dall'implacabile Irina.
